We present a method for parameter estimation in a two-compartment hidden Markov model of the first two stages of hematopoiesis. Hematopoiesis is the specialization of stem cells into mature blood cells. As stem cells are not distinguishable in bone marrow, little is known about their behavior, although it is known that they have the ability to self-renew or to differentiate to more specialized (progenitor) cells. We observe progenitor cells in samples of bone marrow taken from hybrid cats whose cells contain a natural binary marker. With data consisting of the changing proportions of this binary marker over time from several cats, estimates for stem cell self-renewal and differentiation parameters are obtained using an estimating equations approach. 相似文献

Melanocytes are cells of neural crest origin. In the human epidermis, they form a close association with keratinocytes via their dendrites. Melanocytes are well known for their role in skin pigmentation, and their ability to produce and distribute melanin has been studied extensively. One of the factors that regulates melanocytes and skin pigmentation is the locally produced melanocortin peptide alpha-MSH. The effects of alpha-MSH on melanogenesis are mediated via the MC-1R and tyrosinase, the rate-limiting enzyme in the melanogenesis pathway. Binding of alpha-MSH to its receptor increases tyrosinase activity and eumelanin production, which accounts for the skin-darkening effect of alpha-MSH. Other alpha-MSH-related melanocortin peptides, such as ACTH1-17 and desacetylated alpha-MSH, are also agonists at the MC-1R and could regulate melanocyte function. Recent evidence shows that melanocytes have other functions in the skin in addition to their ability to produce melanin. They are able to secrete a wide range of signal molecules, including cytokines, POMC peptides, catecholamines, and NO in response to UV irradiation and other stimuli. Potential targets of these secretory products are keratinocytes, lymphocytes, fibroblasts, mast cells, and endothelial cells, all of which express receptors for these signal molecules. Melanocytes may therefore act as important local regulators of a range of skin cells. It has been shown that alpha-MSH regulates NO production from melanocytes, and it is possible that the melanocortins regulate the release of other signalling molecules from melanocytes. Pathogens causing sexually transmitted diseases (STDs) often consist of related strains that cause non-sexually transmitted, or ''ordinary infectious'', diseases (OIDs). We use differential equation models of single populations to derive conditions under which a genetic variant with one (e.g. sexual) transmission mode can invade and successfully displace a genetic variant with a different (e.g. non-sexual) transmission mode. Invasion by an STD is easier if the equilibrium population size in the presence of an OID is smaller; conversely an OID can invade more easily if the equilibrium size of the population with the STD is larger. Invasion of an STD does not depend on the degree of sterility caused by the infection, but does depend on the added mortality caused by a resident OID. In contrast, the ability of an OID to invade a population at equilibrium with an STD decreases as the degree of sterility caused by the STD increases. When equilibrium population sizes for a population infected with an STD are above the point at which non-sexual contacts exceed sexual contacts (the sexual–social crossover point) and when equilibrium population sizes for an OID are below this point, there can be a stable genetic polymorphism for transmission mode. This is most likely when the STD is mildly sterilizing, and the OID causes low or intermediate levels of added mortality. Because we assume the strains are competitively equivalent and there are no heterogeneities associated with the transmission process, the polymorphism is maintained by density-dependent selection brought about by pathogen effects on population size. 相似文献

In two‐stage group sequential trials with a primary and a secondary endpoint, the overall type I error rate for the primary endpoint is often controlled by an α‐level boundary, such as an O'Brien‐Fleming or Pocock boundary. Following a hierarchical testing sequence, the secondary endpoint is tested only if the primary endpoint achieves statistical significance either at an interim analysis or at the final analysis. To control the type I error rate for the secondary endpoint, this is tested using a Bonferroni procedure or any α‐level group sequential method. In comparison with marginal testing, there is an overall power loss for the test of the secondary endpoint since a claim of a positive result depends on the significance of the primary endpoint in the hierarchical testing sequence. We propose two group sequential testing procedures with improved secondary power: the improved Bonferroni procedure and the improved Pocock procedure. The proposed procedures use the correlation between the interim and final statistics for the secondary endpoint while applying graphical approaches to transfer the significance level from the primary endpoint to the secondary endpoint. The procedures control the familywise error rate (FWER) strongly by construction and this is confirmed via simulation. We also compare the proposed procedures with other commonly used group sequential procedures in terms of control of the FWER and the power of rejecting the secondary hypothesis. Since it was first detected in 1999, West Nile virus (WNV) quickly spread, becoming the dominant vector-borne disease in North
America. Sometimes fatal to humans, WNV is even more widespread among birds, with hundreds of species known to be susceptible
to WNV infection in North America alone. However, despite considerable mortality and local declines observed in American crows
(Corvus brachyrhynchos), there has been little evidence of a large regional association between WNV susceptibility and population declines of any
species. Here we demonstrate a correlation between susceptibility to WNV measured by large-scale testing of dead birds and
two indices of overall population change among bird species following the spread of WNV throughout California. This result
was due primarily to declines in four species of Corvidae, including all species in this family except common ravens (Corvus corax). Our results support the hypothesis that susceptibility to WNV may have negative population consequences to most corvids on
regional levels. They also provide confirmation that dead animal surveillance programs can provide important data indicating
populations most likely to suffer detrimental impacts due to WNV. 相似文献

The simultaneous action of shear deformation and high pressure (SDHP) creates changes in the structure of wood and its main
components (cellulose, hemicelluloses, lignin). The formation of water and alkali soluble polysaccharides under SDHP action,
proceeds in seconds in the solid state, without the use of any reagents and solvents. Therefore, SDHP seems to be a technologically
safe method and friendly to the environment. The amorphization of cellulose crystallites and depolymerization of cellulose
chains were observed under a wide range of pressures (1–6 GPa), both for cellulose samples and the cellulose part of wood.
Similar depolymerization occurs in the hemicellulose part of wood. The decomposition of polysaccharides under SDHP causes
the formation of the water soluble part, whose content increases with pressure and the applied shear deformation. A maximum
solubility of 40% and 55% was registered at 6 GPa following treatment of cellulose and birch wood samples. A higher output
in the case of wood can be explained by a specific role of lignin under SDHP, which acts as a ‘grinding stone’ during cellulose
and hemicelluloses destruction. As shown by high-performance size exclusion chromatography, the water soluble fraction obtained
from cellulose contained glucose (2.6%), cellobiose (9.6%), cellotriose (16.6%) and other higher water soluble oligomers (71%).
Almost complete dissolution (98%) of the treated cellulose sample can be achieved by extraction with 10% NaOH solution. The
SDHP treated birch wood was subjected to submerged fermentation (with Trichoderma viride), and a 13% output of proteins was
obtained. In this case, the water soluble part played the role of the so called ‘start sugars’. Abbreviations: ASF, alkali

Prior to the completion of the Human Genome Project, bioethicists and other academics debated the impact of this new genetic information on medicine, health care, group identification, and peoples’ lives. A major issue is the potential for unintended and intended adverse consequences to groups and individuals. When conducting research in, for instance, American Indian and Alaskan native (AI/AN) populations, political, cultural, religious and historical issues must be considered. Among African Americans, the Tuskegee Syphilis Experiment is a reminder of racism and discrimination in this country. The goal of the current study is to understand reasons for participating, or not, in genetic research such as the HapMap project and other genetic/medical research from the perspective of the Indian American community in Houston, Texas. In this article, we report on a topic central to this discussion among Indian Americans: karma and reincarnation. Both concepts are important beliefs when considering the body and what should happen to it. Karma and reincarnation are also important considerations in participation in medical and genetic research because, according to karma, what is done to the body can affect future existences and the health of future descendants. Such views of genetic and medical research are culturally mediated. Spiritual beliefs about the body, tissue, and fluids and what happens to them when separated from the body can influence ideas about the utility and acceptability of genetic research and thereby affect the recruitment process. Within this community it is understood that genetic and environmental factors contribute to complex diseases such as diabetes, hypertension, and cancer; and acknowledgment of the significance of environmental stressors in the production of disease. A commitment to service, i.e. “betterment of humanity,” karmic beliefs, and targeting environmental stressors could be prominent avenues for public health campaigns in this population. This study suggests that minority status does not automatically indicate unwillingness to participate in genetic or medical research. Indian Americans were not skeptical about the potential benefits of biomedical research in comparison to other ethnic minority communities in the United States. 相似文献

The purpose of a drinking water distribution system is to deliver drinking water to the consumer, preferably with the same quality as when it left the treatment plant. In this context, the maintenance of good microbiological quality is often referred to as biological stability, and the addition of sufficient chlorine residuals is regarded as one way to achieve this. The full-scale drinking water distribution system of Riga (Latvia) was investigated with respect to biological stability in chlorinated drinking water. Flow cytometric (FCM) intact cell concentrations, intracellular adenosine tri-phosphate (ATP), heterotrophic plate counts and residual chlorine measurements were performed to evaluate the drinking water quality and stability at 49 sampling points throughout the distribution network. Cell viability methods were compared and the importance of extracellular ATP measurements was examined as well. FCM intact cell concentrations varied from 5×103 cells mL−1 to 4.66×105 cells mL−1 in the network. While this parameter did not exceed 2.1×104 cells mL−1 in the effluent from any water treatment plant, 50% of all the network samples contained more than 1.06×105 cells mL−1. This indisputably demonstrates biological instability in this particular drinking water distribution system, which was ascribed to a loss of disinfectant residuals and concomitant bacterial growth. The study highlights the potential of using cultivation-independent methods for the assessment of chlorinated water samples. In addition, it underlines the complexity of full-scale drinking water distribution systems, and the resulting challenges to establish the causes of biological instability. 相似文献

MicroRNA-155 has been shown to play a role in immune activation and inflammation, and is suppressed by IL-10, an important anti-inflammatory cytokine. The established involvement of IL-10 in the murine model of Borrelia burgdorferi-induced Lyme arthritis and carditis allowed us to assess the interplay between IL-10 and miR-155 in vivo. As reported previously, Mir155 was highly upregulated in joints from infected severely arthritic B6 Il10-/- mice, but not in mildly arthritic B6 mice. In infected hearts, Mir155 was upregulated in both strains, suggesting a role of miR-155 in Lyme carditis. Using B. burgdorferi-infected B6, Mir155-/-, Il10-/-, and Mir155-/- Il10-/- double-knockout (DKO) mice, we found that anti-inflammatory IL-10 and pro-inflammatory miR-155 have opposite and somewhat compensatory effects on myeloid cell activity, cytokine production, and antibody response. Both IL-10 and miR-155 were required for suppression of Lyme carditis. Infected Mir155-/- mice developed moderate/severe carditis, had higher B. burgdorferi numbers, and had reduced Th1 cytokine expression in hearts. In contrast, while Il10-/- and DKO mice also developed severe carditis, hearts had reduced bacterial numbers and elevated Th1 and innate cytokine expression. Surprisingly, miR-155 had little effect on Lyme arthritis. These results show that antagonistic interplay between IL-10 and miR-155 is required to balance host defense and immune activation in vivo, and this balance is particularly important for suppression of Lyme carditis. These results also highlight tissue-specific differences in Lyme arthritis and carditis pathogenesis, and reveal the importance of IL-10-mediated regulation of miR-155 in maintaining healthy immunity. 相似文献

To determine whether genetic differences in fitness components exist among seeds and seedlings in a natural population, weighed propagules of six parents of Anthoxanthum odoratum from a reciprocal diallel cross were planted into the parental source population, a mown field. Seed families of maternal genotypes differed in germination success, while paternal families showed no detectable differences. Differential germination success could not be attributed to propagule weight. Seed families ranked differently in germination percentage in different blocks. No survivorship differences among parental seed families could be detected. There were significant cross × block × germination and cross × block × survivorship interactions; different crosses performed better or worse in different blocks. In some cases, crosses sired by different fathers within a maternal seed family differed in germination or survivorship, suggesting that natural selection may be capable of discriminating among juvenile genotypes within a maternal family despite the presence of large overall maternal effects. These results indicate that seedling establishment may differ according to genotype and that microsite heterogeneity may maintain genetic variation in juvenile traits in natural plant populations. 相似文献
